Cell viability reagent (AO/DAPI)

for ADAM™ MC Plus & CellT Plus

Cell viability reagent is a single reagent composed of mixture of Acridine Orange and 4′,6-diamidino-2-phenylindole. It is to be used with AccuPlus Slide, the disposable 4 channel slide for ADAM MC Plus & CellT Plus.

Cell viability reagent is to be used with AccuPlus Slide, which is a disposable slide for ADAM MC Plus & CellT Plus


Measuring process is much simple and convenient with this single reagent composed of the mixture of AO and DAPI.


It is possible to accurately measure not only the numbers of total cells, viable cells, non-viable cells, and viability, 

but also the cell size and cell aggregation ratio by using an advanced algorithm of cell counting and analysis. 






 


Staining method

 

AO (Acridine Orange) is a permeable dye that stains the nucleus of every cell regardless of the cell’s condition. 

Therefore, it is used to count the total number of cells. 


DAPI (4′,6-diamidino-2-phenylindole) is an impermeable dye that only stains the nucleus of cells with damaged membranes or cells with inactive metabolism. 

Therefore, cells with nucleus stained with DAPI are counted as non-viable cell. 


After taking fluorescent images of dyed samples, obtained images are processed by image analysis software integrated in the system. 

Finally, the total cell concentration and cell viability of the sample are automatically calculated and displayed on the screen, 

after each measurement of the total cells and the non-viable cells.
